Hydrothermal treatment to enhance supercritical CO2 poly-caprolactone foaming processes for tissue engineering scaffolds

Abstract
Se ha estudiado el tratamiento hidrotérmico como estrategia para mejorar el proceso de espumado o foaming con CO2 supercrítico para la fabricación de scaffolds (o andamios) de policaprolactona (PCL) destinados a aplicaciones de ingeniería tisular. Las muestras de PCL se han sometido a foaming supercrítico a 300 bar y 40 °C durante 60 minutos, combinado con tratamientos hidrotérmicos realizados antes o después del espumado a temperaturas de 70-100 °C y presiones de 10-20 bar. Los efectos de estos tratamientos sobre la morfología, la porosidad y el comportamiento mecánico de los andamios se evaluaron mediante microscopía electrónica de barrido, microtomografía computarizada y ensayos de compresión. Los resultados mostraron que el tratamiento hidrotérmico previo al espumado mejoraba significativamente la porosidad del scaffold al tiempo que aumentaba la interconectividad de los poros. Por el contrario, el tratamiento hidrotérmico posterior al espumado provocó el colapso de los poros y la pérdida de integridad estructural. Estos hallazgos demuestran que la combinación del pretratamiento hidrotérmico con el espumado con CO2 supercrítico proporciona una vía sostenible y libre de disolventes para adaptar la morfología y el rendimiento mecánico de los scaffolds generados.
 
: Hydrothermal treatment has been explored as a strategy to enhance the supercritical CO₂ 11 foaming process for fabricating polycaprolactone (PCL) scaffolds aimed at tissue engineering appli12 cations. This study investigates the effect of hydrothermal treatment applied before and after the 13 foaming process on the morphological and mechanical properties of PCL scaffolds. PCL samples 14 underwent supercritical CO₂ foaming followed by hydrothermal treatment under different condi15 tions. Characterization techniques revealed that pre-foaming hydrothermal treatment improved 16 scaffold porosity, pore size distribution, and interconnectivity, while post-foaming treatment led to 17 structural compromise. Mechanical evaluation confirmed that pre-treated scaffolds maintained 18 their integrity, whereas post-treated scaffolds exhibited excessive fragility. Additionally, the study 19 underscores the importance of selecting optimal processing conditions to balance porosity and me20 chanical properties. These findings highlight the potential of combining hydrothermal treatment 21 with supercritical CO₂ foaming to optimize scaffold design, offering a solvent-free approach for tai22 loring porosity and mechanical properties. Future studies should focus on biological evaluations, 23 including in vitro cell culture and in vivo testing, to further validate their applicability in regenera24 tive medicine and tissue engineering. Further optimization of processing conditions could lead to 25 scaffolds with enhanced bioactivity and controlled degradation rates.
