RT journal article T1 Plasma 1α-Hydroxycorticosterone as Biomarker for Acute Stress in Catsharks (Scyliorhinus canicula) A1 Ruiz Jarabo de la Rocha, Ignacio A1 Barragan-Mendez, Cristina A1 Jerez Cepa, Ismael A1 Fernández-Castro, Miriam A1 Sobrino, Ignacio A1 Mancera Romero, Juan Miguel A1 Aerts, Johan K1 1a-hydroxycorticosterone K1 glucocorticoid K1 Scyliorhinus canicula K1 shark K1 stress AB Glucocorticoids are pleiotropic steroid hormones mediating redistribution of energy. They induce breakdown of glycogen stores and consequent plasma hyperglycaemiaafter stressful situations. Glucocorticoid actions in most vertebrate species areexerted by cortisol and corticosterone. However, 1a-hydroxycorticosterone is thedominant corticosteroid hormone in elasmobranchs, though its effects as aglucocorticoid are unknown. Here we demonstrate, by using ultra-performanceliquid chromatography coupled to tandem mass spectrometry for the quantificationof 1a-hydroxycorticosterone in plasma of the elasmobranch Scyliorhinus canicula, the response of this hormone to an acute-stress situation and for the first timeits glucocorticoid action in elasmobranchs. After an acute air-exposure challenge,S. canicula increased plasma levels of 1a-hydroxycorticosterone altogether withenhanced glycolysis and gluconeogenesis pathways to fuel energy demanding tissues, such as white muscle, during the first hours after the stress situation. We foresee ourstudy as a starting point to evaluate stress responses in elasmobranchs, as well as forfuture applications in the management of these key ecosystem species.
