Abstract

The need to establish sustainable aquaculture is driving a shift towards replacing fishmeal with plant-based alternatives such as soya, which requires nutritional assessment tools that are efficient, cost-effective and ethically sound. The main aim of this study was to design and validate an in vitro digestion model for the lemon seabream (Seriola dumerili). To this end, free amino acids were measured using the OPA method, employing enzymatic extracts. The effect of the acid phase was evaluated by comparing a sequential model (acid and alkaline phases) with a single-phase model (direct alkaline phase), under temperature gradients of 18°C, 22°C and 26°C, using fish meal and soya meal as protein sources. The results indicated that the inclusion of the acid phase in digestion exerts a synergistic effect that maximises the efficiency of intestinal enzymes in the subsequent alkaline phase, also showing that fish meal concentrates 95% of its release in the alkaline phase, whilst soya meal exhibits higher release values in the acid phase. Regarding temperature dependence, fish meal showed a linear increase in amino group release as a function of temperature, whilst soya meal remained stable with increasing temperature. Furthermore, the trials revealed that at 18°C thermodynamic limitations reduce the rate of hydrolysis, at 26°C there is a sharp increase in activity in single-phase digestion, and 22°C was established as the point of maximum resolution and sensitivity of the model.
